摘要:
目的观察闽产三叶青地上部分抗炎提取物的小鼠急性毒性反应,初步评价其用药安全性。方法取昆明小鼠60 只,雌雄各半,按体质量随机分为各给药组及空白组;给药组按0.3 mL/10 g最大给药体积,以0.75 g/mL闽产三叶青地上部分抗炎提取物(THAA)的最大浓度药液、0.67 g/mL闽产三叶青粗提物(粗提物)的最大浓度药液分组给药;空白组灌服0.3 mL/10 g蒸馏水。24 h内灌胃给药 3 次,间隔8 h,一次性口服灌胃给药,连续观察14 d,观察其急性毒性症状谱,记录小鼠累计死亡数及小鼠体质量变化。结果THAA、粗提物均无法得出半数致死量。观察期间各组小鼠未见中毒反应,无动物死亡;给药14 d后解剖肉眼未见脏器异常;空白组与各给药组间的脏器指数评价及小鼠体质量变化比较,差别无统计学意义(P<005)。结论THAA及粗提物的小鼠灌胃给药最大耐受剂量分别为90.0,80.4 g·kg-1·d-1,分别相当于临床60 kg人每公斤体质量日用量的360.0倍及321.6倍。THAA及THAA粗提物口服毒性小,临床剂量下用药安全可靠。
Abstract:
ABSTRACT:ObjectiveTo observe the acute toxicity of antiinflammatory extracts of Tetrastigmatis hemsleyanum’s aerial part in mice, and evaluate the drug safety.Methods60 Kunming mice, half male and half female, were randomly divided into drug groups and blank group according to body mass.The drug groups received antiinflammatory extracts of Tetrastigmatis hemsleyanum’s aerial part by gavage for 3 times a day, interval of 8 hours, and the volume of administration was 0.3 mL/10 g, and the concentration of administration was 0.67 g/mL of antiinflammatory extracts of Tetrastigmatis hemsleyanum’s aerial part and 0.75 g/mL of crude extracts.The blank group received distilled water, and the volume of administration was 0.3 mL/10 g.For 14 days, the toxic reaction and death number of mice were recorded.ResultsThe LD50 can’t be drawn in antiinflammatory extracts of Tetrastigmatis hemsleyanum’s aerial part and crude extracts, during the 14 days, toxic reaction and death were not appeared in mice.After 14 days, abnormal viscera were not found by naked eye after anatomy, and there was no significant difference in evaluation of viscera index and mouse weight change between drug group and blank group.ConclusionThe maximal tolerance dose of antiinflammatory extracts of Tetrastigmatis hemsleyanum’s aerial part and crude extracts are 90.0 g·kg-1·d-1 and 80.4 g·kg-1·d-1 in mice, respectively, which is equal to 321.6 times and 360.0 times of human (weighted 60 kg) daily dried medicinal herb expenses.The oral toxicity of antiinflammatory extracts of Tetrastigmatis hemsleyanum’s aerial part and crude extracts is small, and its clinical dosage is safe and reliable.
