摘要:
目的评估长链非编码RNA(lncRNA) INK4位点的反义非编码RNA(ANRIL)在银屑病患者病变皮肤组织中的表达水平及其与疾病严重程度和炎症因子的关联。方法连续纳入204例银屑病患者,钻取银屑病病变组织和正常皮肤组织标本,采用荧光定量聚合酶链反应检测病变组织及配对正常皮肤组织的lncRNA ANRIL以及炎症因子的表达量。采用银屑病皮损面积及严重度指数(PASI)评估银屑病的严重程度。结果lncRNA ANRIL在病变皮肤组织中的表达量高于配对正常皮肤组织(P<0.001)。受试者工作特征曲线显示,lncRNA ANRIL表达水平可以较好地区分银屑病病变皮肤组织和配对正常皮肤组织(曲线下面积:0.711,95%CI:0.656~0.766)。患者病变皮肤组织的lncRNA ANRIL表达量与PASI评分正相关(r=0.261,P<0.001)。进一步行多元线性回归分析,结果显示,病变皮肤组织lncRNA ANRIL高表达(P=0.013)、受累体表面积较大(P<0.001)和同时接受局部治疗(P=0.008)是患者较高PASI评分的独立相关因素。此外,病变皮肤组织的lncRNA ANRIL表达量与促炎因子肿瘤坏死因子α(TNF|α)(r=0.248, P<0.001)、白细胞介素|6(IL|6)(r=0.219, P=0.002)和IL|17(r=0.252, P<0.001)的表达水平正相关。结论lncRNA ANRIL在银屑病病变皮肤组织中高表达,且与患者的疾病严重程度和炎症因子水平正相关。
Abstract:
ObjectiveTo investigate the expression of long non|coding RNA (lncRNA) antisense noncoding RNA in the INK4 locus (ANRIL) in psoriatic lesioned skin tissue of psoriasis patients, and its association with disease severity as well as inflammatory cytokines levels.Methods204 psoriasis patients were consecutively enrolled, and 3 mm deep psoriatic lesioned skin tissue and paired normal skin tissue 5 cm away from the lesion were collected for the detection of lncRNA ANRIL expression and inflammatory cytokines levels by quantitative polymerase chain reaction.ResultslncRNA ANRIL expression in lesioned skin tissue was higher than that in paired normal skin tissues (P<0.001).Receiver operating characteristic curve showed that the area under the curve of lncRNA ANRIL expression for distinguishing psoriasis lesions from paired normal skin tissue was 0.711(95% CI:0.656|0.766).lncRNA ANRIL expression in lesioned skin tissue was positively correlated with PASI score (r=0.261, P<0.001).And high expression of lncRNA ANRIL in lesioned skin tissue (P=0.013), large BAS involvement (P<0.001) as well as simultaneous local treatment (P=0.008) were independently associated with higher PASI score.In addition, lncRNA ANRIL expression level in lesion skin tissue was positively associated with pro|inflammatory cytokines tumor necrosis factor|α (r=0.248, P<0.001), interleukin (IL)|6 (r=0.219, P=0.002) and IL|17 (r=0.252, P<0.001) levels.ConclusionslncRNA ANRIL is highly expressed in lesioned skin tissue and is positively correlated with disease severity and inflammatory cytokines levels in patients with psoriasis.
