摘要:
目的 评估长链非编码RNA 核富集转录本1(lncRNA NEAT1)在银屑病患者病变皮肤组织中的表达水平及其与疾病严重程度和炎症因子水平的关联。 方法 连续纳入204例银屑病患者,钻取3 mm深度银屑病病变组织标本,并同时钻取病变5 cm外的配对正常皮肤组织标本。采用定量聚合酶链反应检测患者病变组织及配对正常皮肤组织的lncRNA NEAT1表达量和病变组织标本的肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、IL-8、IL-17和IL-22的mRNAs表达量。采用银屑病皮损面积及严重度指数(PASI)评估银屑病严重程度。 结果与配对正常皮肤组织比较,lncRNA NEAT1在病变组织中的表达量明显升高(P<0.001)。ROC曲线结果显示,lncRNA NEAT1表达量可以较好地区分病变皮肤组织和正常组织(曲线下面积为0.747,95%CI:0.700~0.794)。此外,病变皮肤组织lncRNA NEAT1表达量与PASI评分呈正相关(r=0.275,P<0.001),而与其他临床特征无关(均P>0.05)。进一步采用多元线性回归模型分析显示,病变皮肤组织lncRNA NEAT1表达量(P=0.014)、受累BSA(P<0.001)和局部治疗(P=0.008)均与PASI呈独立正相关。病变皮肤组织lncRNA NEAT1表达量与TNF-α(r=0.342, P<0.001)、IL-6(r=0.315, P<0.001)、IL-8(r=0.174, P=0.013)、IL-17(r=0.345, P<0.001)和IL-22(r=0.146, P=0.037)的mRNAs表达量均呈正相关。 结论 lncRNA NEAT1在银屑病患者病变皮肤组织中高表达,且与疾病活动度和炎症因子表达量呈正相关。
Abstract:
Objective To assess the expression of long non-coding RNA nuclear enriched abundant transcript 1(lncRNA NEAT1)and its correlations with disease activity as well as inflammatory factors levels in psoriasis patients. Methods 204 psoriasis patients were consecutively enrolled in this study.lncRNA NEAT1 expression was detected in normal tissue and psoriasis affected area by quantitative polymerase chain reaction(qPCR). Inflammatory factors mRNA levels(including tumor necrosis factor-α (TNF-α), interleukin(IL)-6(IL-6), IL-8, IL-17 and IL-22)in psoriasis affected area were also detected by qPCR. Results Compared to paired normal tissue, lncRNA NEAT1 was highly expressed in psoriasis affected area(P<0.001). Receiver operating characteristic(ROC)curve disclosed that lncRNA NEAT1 expression could well distinguish psoriasis affected area from paired normal tissue(area under the curve), 0.747, 95%CI:0.700-0.794). Also, lncRNA NEAT1 expression was positively correlated with psoriasis area and severity index(PASI)score(r=0.275, P<0.001). Multivariant regression analysis further revealed that increased lncRNA NEAT1 expression(P=0.014), larger BSA(P<0.001)and local treatment(P=0.008)were correlated with PASI score independently. Moreover, lncRNA NEAT1 expression was positively associated with Inflammatory factors mRNA levels(including tumor necrosis factor-α(TNF-α)(r=0.342, P<0.001), interleukin(IL)-6(IL-6)(r=0.315, P<0.001), IL-8(r=0.174, P=0.013), IL-17(r=0.345, P<0.001)and IL-22(r=0.146, P=0.037)). Conclusion lncRNA NEAT1 expression was highly expressed in psoriasis affected tissue and positively correlated with disease activity as well as inflammatory factors levels in psoriasis patients.
