摘要:
目的检测miR-581在食管鳞状细胞癌组织中的表达情况及对食管鳞癌细胞K30增殖、迁移和侵袭能力的影响。方法收集33例食管鳞状细胞癌组织及其癌旁组织标本,应用实时荧光定量PCR(qPCR)方法检测miR-581的表达情况; 采用miR-581 mimic转染食管鳞癌细胞K30作为实验组,用阴性对照片段转染K30作为阴性对照组,qPCR检测miR-581的转染效率,CCK8检测细胞增殖能力的变化,Transwell实验检测迁移和侵袭能力的变化。结果与食管癌旁组织比较,癌组织中的miR-581表达下调; CCK8结果显示,过表达miR-581能抑[JP2]制食管癌细胞的增殖能力; Transwell迁移及侵袭实验结果显示,过表达miR-581能显著减少食管癌细胞穿膜细胞数。结论miR-581与食管鳞癌的进展相关,其机制有待进一步研究。
Abstract:
ObjectiveTo detect the expression of miR-581 in esophageal squamous cell carcinoma(ESCC)tissues and investigate the effect of miR-581 on the proliferation, migration and invasion of K30 cells.MethodsThe expression levels of miR-581 in ESCC tissues and ESCC cells were detected by qPCR.The miR-581 mimic and negative control were transfected into ESCC K30 cells using Lipofectamine 2000.The cell proliferation was determined by CCK8 assay.The effect of miR-581 on the migration and invasion was analyzed by transwell assay.ResultsCompared with the adjacent normal tissues, the expression level of miR-581 was remarkably decreased in the ESCC tissues.After transected with miR-581 mimic, the capability of proliferation, migration and invasion in K30 cells were obviously inhibited.ConclusionmiR-581 may be associated with the development of ESCC while the underlying mechanism needs to be further explored.
